Final Report Abstract

The expression of genes related to symbiotic nitrogen fixation (Lb A, Lb B, Fd, Sst1) were investigated in Pisum sativum under low and high levels of sulfur deficiency during the vegetative and generative stage. Total biomass, shoot dry weight, nodule quantity, and total RNA extracted from leaf and nodule are significantly lowered under severe sulfur deficiency. Chlorophyll content was reduced due to sulfur deficiency and after the switch from the vegetative to the generative stage. In nodules obtained during the generative phase, expression of both, Lb A and Lb B genes decreased significantly under high sulfur deficiency vs. control. In contrary, the expression was higher during the vegetative stage under the same condition. The expression of Fd in the nodules increased at high sulfur deficiency compared to the control and decreased during plant development. In leaf and root tissues, Fd expression in vegetative phase was higher compared to the generative phase. It decreased under high sulfur deficiency vs. control. Sst1 was identified as a sulfate transporter in nodules of Pisum sativum . By use of the Saccharomyces cerevisiae mutant strainCP154-7A, the sulfate transporter function was verified. mRNA abundance of Sst1 was reduced at high sulfur deficiency vs. control.
