Genome-wide Transcriptional Programs Regulating Invasive Growth of Human Breast and Brain Cancer

Two of the most aggressive types of human cancer, glioblastoma and breast cancer, are driven by a shared biochemical network comprising growth factors of the EGF family and their cognate receptors, the EGFR/HER2 tyrosine kinases. Currently available evidence suggests that late stages of tumor progression and invasive growth are propelled by genome-wide transcriptional programs, which are instigated by growth factors and tumor environment. Hence, we aim at an unprecedentedly exhaustive transcriptomic landscape of 3D model cultures, in which the network will be stimulated either transiently (by growth factors) or permanently (by oncogenic mutations). High throughput platforms available at the German Cancer Research Center will enable us to derive whole genome mRNA data, all alternatively spliced isoforms, as well as information on most of the existing micro-RNAs and large intervening non-coding RNAs. The data will be integrated by using a package of software analyses developed at the Department of Physics of Complex Systems of the Weizmann Institute. Predictably, the emerging picture will converge on specific modules, whose contribution to late-stage invasive growth and poor prognosis of cancer patients will be determined at the University Clinics Heidelberg. By interfacing model systems with clinical data, our collaborative studies will identify new tumor predictors and bio-markers, as well as establish novel informatics and other tools capable of voluminous data integration and pathway prediction In the context of disease.
