The C3 work program addresses four topics: (i) the mechanistic characterization of HCMV-specific core NEC heterodimerization, (ii) the question of conservation of this essential NEC scaffolding principle among herpesviruses, (iii) a detailed analysis of the various regulatory functions recruited to the multimeric NEC and, (iv) an exploitation of the NEC as a putative cross-spanning antiherpesviral drug target. Thus, our project will provide new information on the NEC-specific structure-function relationship focusing on its multifaceted mode of virus-host interaction.
