TNF is a critical pathogenic factor in psoriasis and other chronic inflammatory diseases, but the mechanisms by which TNF triggers inflammation remain poorly understood. TNFR1 signalling induces inflammatory gene expression by activating the IKK/NF-kB pathway, but also triggers cell death by inducing apoptosis and necroptosis. We use genetic mouse models to study the role and contribution of IKK/NF-kB signalling but also of apoptosis and necroptosis in the TNF-mediated mechanisms regulating skin homeostasis and inflammation.
